Off-Hand I would have said Wonder Woman #1 since she has a rather patriotic outfit and is shown charging into obviously German soldiers. However, The Shield was the first bonafide "patriotic" super hero.

Outside the superhero genre I imagine there were probably patriotic covers predating both.

I'd guess there migth be an early New Fun comics with a 4th of July theme.
